| Synthesis conditions:Prior to dilution ensure that all product is at the bottom of the vial. Dilute to the recommended concentration and mix thoroughly in the sealed vial to ensure that all contents are dissolved. For best results an extra 5 minute capping step should be included after the final oxidation step. This may be done on the synthesizer or by syringe flushing the column.A mixture of 1 mL CAP A (Acetic anhydride/THF 1:8) and 1 mL of CAP B (N-methyl imidazole/THF 1:8) should be used to saturate the column for 5 minutes.The column should be washed with dry acetonitrile after the capping step. The extra capping step will eliminate a byproduct that elutes after the product under AX HPLC analysis. We recommend using ethyl thiol tetrazole for the activator reagent with this product.Dilution:100 mg/mLCoupling:10-minute couplingDeprotection conditions:When using fast deprotecting amidites (eg. C-Ac, G-DMF, G-PAC) use t-butylamine/Water (1:3) for 6 hours at 60 °C. When using standard amidites (eg. C-Bz, G-iBu) use t-butylamine/Water (1:3) overnight (15-24 hours) at 60 °C.
