| T7 R&DNA™ Polymerase and the correspond ing wild-type T7 RNA polymerase are useful for invitro synthesis of defined “transcripts” that are complementary to nucleic acids cloned into a plasmid or other vector downstream from a T7 RNA polymerase promoter. In contrast to the corresponding wild-type T7 RNA polymerase, the T7 R&DNA Polymerase can incorporate 2′-deoxy ribonucleoside-5′-triphosphates or other 2′-modi fied triphosphates such as ribonuclease-resis tant 2′-fluoro-ribonucleoside-5′-triphosphates, in addition to the canonical ribonucleoside-5′-tri phosphates. The ability of this mutant enzyme to incorporate various non-canonical 2′-ribonu cleotides permits either primed or unprimed invitro synthesis of “transcripts” composed of rNMPs, dNMPs, modified 2′-NMPs, or of mixed dNMP/rNMP, or 2′-modified-NMP/rNMP composi tion for a variety of applications. 该酶是T7 RNA聚合酶的突变形式(Y639F突变体)。该突变使得T7 R&DNA聚合酶能够比相应的野生型T7 RNA聚合酶更有效地将2’-脱氧核糖核苷三磷酸(dNTP)掺入全长“RNA”转录物中。该突变酶使用与野生型T7 RNA聚合酶相同的T7转录启动子。 |
