This column, packed with Biosearch Technologies' 5'-DMT-mdC(TEG-ROX)-Phosphate CPG, allows for the introduction of a reporter molecule onto the 3'-terminus end of the oligonucleotide. 5'-DMT-mdC(TEG-ROX)-Phosphate CPG is based on a modified cytosine residue linked to the ROX moiety via a triethyleneglycol spacer. The 3' end is conjugated to the CPG support via phosphate linkage.

产品用途:
Synthesis conditions:Follow the instrument manufacturer's protocol, with noted changes, when using this product. Synthesis can be performed on these supports according to standard procedures with deprotection and cleavage modifications noted.Cleavage conditions:Use t-butylamine/water (1:3) for 45 minutes at 25 °C. The use of ammonia must be avoided, as it will cause degradation of the ROX moiety.Deprotection conditions:When using fast deprotecting amidites (eg. C-Ac, G-DMF, G-PAC) use t-butylamine/Water (1:3) for 6 hours at 65° C. When using standard amidites (eg. C-Bz, G-iBu) use t-butylamine/Water (1:3) overnight (12-15 hours) at 65°C. The use of ammonia must be avoided, as it will cause degradation of the ROX moiety. Image of cleaved and deprotected structure:
