产品详细信息Description: The monoclonal antibody MH9A4 recognizes interleukin-9 (IL-9), a proinflammatory cytokine historically believed to be involved in type 2 immune responses. Recent evidence suggests IL-9 may instead be secreted by a distinct T-helper lineage called Th9. These cells can be derived from Th2 cells with TGF beta or differentiated directly from naive CD4+ T cells with TGF beta and IL-4.IL-9 is a member of the common cytokine receptor gamma chain-dependent family of cytokines whichalso includes IL-2, IL-4, IL-7, IL-15 and IL-21. Its pleiotropic effects on Th2 lymphocytes, B lymphocytes, mast cells, eosinophils and gut and airway epithelial cells have implicated IL-9 in asthma and other allergy-related disorders. Staining with the MH9A4 antibody is not blocked by the MH9D1 antibody, suggesting that the two antibodies bind to unique epitopes.Applications Reported: This MH9A4 antibody has been reported for use in intracellular staining followed by flow cytometric analysis.Applications Tested: This MH9A4 antibody has been pre-titrated and tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells. This can be used at 5 µL (0.06 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.eFluor® 660 is a replacement for Alexa Fluor® 647. eFluor® 660 emits at 659 nm and is excited with the red laser (633 nm). IL-9 regulates many cell types involved in classical Th2-associated asthma responses, including B and T lymphocytes, mast cells, eosinophils, and epithelial cells. The mouse IL-9 cDNA encodes a 144 amino acid precursor protein with an 18 amino acid signal peptide that is cleaved to form the mature cysteine-rich protein with a molecular mass of 14 kDa. Native mouse IL-9 is heavily glycosylated. Recently, TGF beta has beenreported to play an important role in the differentiation of IL-17-producing T helper cells and inducible regulatory T cells by reprogramming Th2 cells to secrete IL-9, thereby driving the differentiation of Th9 cells directly.

| IL-9 Antibody (50-7097-42) in FlowTh9-polarized human peripheral blood cells were cultured for 5 hours with Protein Transport Inhibitor Cocktail (Product # 00-4980-03) (left) or were stimulated with Cell Stimulation Cocktail (plus protein transport inhibitors) (Product # 00-4975-03) (right). Cells were then stained intracellularly with Anti-Human CD4 FITC (Product # 11-0049-42) and Anti-Human IL-9 eFluor® 660. Cells were fixed and permeabilized with IC Fixation Buffer (Product # 00-8222-49) and Permeabilization Buffer (10X) (Product # 00-8333-56). Cells in the lymphocyte gate were used for analysis. |
