产品详细信息Description: E-Selectin (CL-37) is a mouse monoclonal antibody raised against E-Selectinof human origin.Endothelial Leukocyte Adhesion Molecule-1 belongs to the selectin family of adhesion molecules. Together with L-selectin and P-selectin, E-selectin mediates the initial interactions of leukocytes and platelets with endothelial cells.Molecular structure: The extracellular part of all selectins consists of an aminoterminal c-type lectin domain which specifically binds to carbohydrate ligands. This is followed by an EGF-like domain, and, in the case of E-selectin, by 6 short consensus repeats. The transmembrane portion of the molecule is followed by a short cytoplasmic tail.Selectins guide non-activated polymorphonuclear cells to the areas of inflammation in creating first, loose contacts with the endothelial layer. Together with P-selectin, E-selectin is expressed on cytokine-activated endothelial cells, and contributes to the adhesion of still resting leukocytes to the endothelium. This initial binding event is an essential prerequisite for the activation of the immune cells via different inflammatory mediators. In contrast to P-selectin, E-selectin is maximally expressed 2-4 hours after cell activation. Within the next 24-48 hours E-selectin is again eliminated from the cytoplasmic membrane by shedding into the circulation.Applications Tested: ELISA, Immunohistochemistry (Frozen).Source: Plant expressed 130 amino acids (His25-Ser153) with 16 amino acid Histidine tag. accession number NP_000580.Bioactivity: The ED50 of this protein, as measured by TF-1 cell proliferation assay, is less than or equal to 0.7 ng/mL. Purity: Greater than or equal to 95% as determined by SDS-PAGE.Molecular Weight: 17.2 kDa (apparent 23 and 24 kDa).靶标信息Leukocytes adhere to the blood vessel endothelium during extravasation in postcapillary venules of lymph nodes. In addition, leukocyte adhesion occurs in the capillaries and small venules at any site in the body after onset of inflammation. This response is immediate and involves a cascade of adhesion receptors. At the endothelial surface members of the selectin and immunoglobulin superfamilies participate in this cascade. Selectins are C-type cell surface lectins that play a role in leukocyte adhesion to the blood vessel wall endothelium. E-selectin (CD62E) is an endothelial cell specific selectin that is expressed only after activation with proinflammatory cytokines. In vitro experiments have shown that IL-1, TNFalpha and bacterial wall components like lipopolysaccharides induce the transcription of E-selectin in a NFkB dependent signaling cascade. E-selectin has been associated with blood vessel endothelium in diverse inflammatory situations.
