靶标信息Histone H3 is one of the DNA-binding proteins found in the chromatin of all eukaryotic cells. H3 along with four core histone proteins binds to DNA forming the structure of the nucleosome. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. Post translationally, histones are modified in a variety of ways to either directly change the chromatin structure or allow for the binding of specific transcription factors. The N-terminal tail ofhistone H3 protrudes from the globular nucleosome core and can undergo several different types of post-translational modification that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

H3K27me2 Antibody (MA5-23511) in WBWestern blot analysis was performed on acid extracts (20 µg lysate) of HeLa (Lane 1), MCF7 (Lane 2), MDA-MB-231 (Lane 3), HCT 116 (Lane 4) and U-2 OS (Lane 5). The blot was probed with Di-Tri-Methyl-Histone H3 (Lys27) (Product # MA5-23511, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 17 kDa band corresponding to Di-Tri-Methyl-Histone H3 (Lys27) was observed across the cell lines tested. H3K27me2 Antibody (MA5-23511) in ARRAYA Dot Blot analysis was performed to test the cross reactivity of Di-Tri-Methyl-Histone H3 (Lys27) monoclonal antibody (Product # MA5-23511) with peptides containing other modifications of histone H3. Other modifications include mono-methylation of lysine 27 and mono-, di- and trimethylation of lysines 9 and 36. One hundred to 0.2 pmol of the peptide containing the respective histone modification were spotted on a membrane. The antibody was used at a dilution of 1:300. |
