产品详细信息This antibody is predicted to react with mouse and rat based on sequence homology.靶标信息Histone H3 is one of the DNA-binding proteins found in the chromatin of all eukaryotic cells. H3 along with four core histone proteins binds to DNA forming the structure of the nucleosome. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. Post translationally, histones are modified in a variety of ways to either directly change the chromatin structure or allow for the binding of specific transcription factors. The N-terminal tail ofhistone H3 protrudes from the globular nucleosome core and can undergo several different types of post-translational modification that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

Phospho-Histone H3 (Thr32) Antibody (PA5-40141) in WBWestern blot analysis was performed on acid extracts (20 µg lysate) of HeLa (Lane 1), HeLa treated with Calyculin A (50 ng/mL for 30 minutes) (Lane 2), RAW264.7 (Lane 3), RAW264.7 treated with Calyculin A (50 ng/mL for 30 minutes) (Lane 4), NIH/3T3 (Lane 5) and NIH/3T3 treated with Calyculin A (50 ng/mL for 30 minutes) (Lane 6). The blot was probed with Anti-Phospho-Histone H3 (Thr32) (Product # PA5-40141, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 17 kDa band corresponding to Phospho-Histone H3 (Thr32) was observed across the cell lines upon treatment.
