Histone H3 is one of the DNA-binding proteins found in the chromatin of all eukaryotic cells. H3 along with four core histone proteins binds to DNA forming the structure of the nucleosome. Histones play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. Post translationally, histones are modified in a variety of ways to either directly change the chromatin structure or allow for the binding of specific transcription factors. The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of post-translational modification that influence cellular processes. These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.

H3K27me1 Antibody (49-1012) in IFHuman osteosarcoma (U2OS) cells were stained with the anti-H3K27me1 antibody (Product # 49-1012) and with DAPI. Cells were fixed with 4% formaldehyde for 20’ and blocked with PBS/TX-100 containing 5% normal goat serum. Figure A: cells were immunofluorescently labeled with the H3K27me1 antibody (left) diluted 1:1,000 in blocking solution followed by an anti-rabbit antibody conjugated to Alexa568 or with DAPI (right), which specifically labels DNA. Figure B, C, D and E: staining of the cells with the H3K27me1 antibody after incubation of the antibody with 2 ng/μL blocking peptide containing the unmodified and the mono-, di- and trimethylated H3K27, respectively. H3K27me1 Antibody (49-1012) in WB Western blot analysis was performed on acid extracts (20 µg lysate) of HeLa (Lane 1), K-562 (Lane 2), MDA-MB-231 (Lane 3) and COS-7 (Lane 4). The blot was probed with Anti-Methyl-Histone H3 (Lys27) Polyclonal Antibody (Product # 49-1012, 1:500 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 17 kDa band corresponding to Methyl-Histone H3 (Lys27) was observed across the cell lines tested. |
