Tetramethylrhodamine CE phosphoramidite (TAMRA Amidite) is used for the labeling of oligonucleotides and fluorogenic probes. This product is prepared using the pure 6-carboxytetramethylrhodamine isomer and can be added only at the 5' terminus of the oligo, thereby terminating the synthesis. For best result we recommend a 10 minute coupling routine and an additional capping step after oxidation. Rhodamine dyes are susceptible to degradation by strong bases such as concentrated NH4OH, we suggest using t-butylamine/H2O for deprotection and cleavage. |

Synthesis conditions:Prior to dilution ensure that all product is at the bottom of the vial. Dilute to the recommended concentration and mix thoroughly in the sealed vial to ensure that all contents are dissolved. Product should only be used for DNA synthesis with standard reagents ( i.e. Ethylthio Tetrazole, 0.02M Iodoine/THF).Dilution:100 µmol/mLCoupling:10-minute couplingDeprotection conditions:When using fast deprotecting amidites (eg. C-Ac, G-DMF, G-PAC) use t-butylamine/Water (1:3) for 6 hours at 65°C. When using standard amidites (eg. C-Bz, G-iBu) use t-butylamine/Water (1:3) overnight (12-15 hours) at 65°C.
